内分泌治疗后前列腺癌上皮间质转化标志物的变化

摘    要:目的探讨内分泌治疗后前列腺癌患者肿瘤组织中上皮间质转化(EMT)标志物表达的变化。方法收集山西医科大学第一医院2006年1月至2012年12月收治的17例经过内分泌治疗前后的前列腺癌患者肿瘤组织标本,采用免疫组织化学链霉亲和素-生物素复合物(SABC)法检测上皮钙黏素(E Cadherin),波形蛋白(Vimentin)、神经钙黏素(N Cadherin)等分子标志物的表达。结果经过内分泌治疗后,前列腺癌患者肿瘤组织中上皮细胞标志E Cadherin表达阳性率由(87.8±5.4)%下降到(6.3±1.7)%,间充质细胞标志N Cadherin表达阳性率由(1.2±0.6)%增加到(46.1±5.6)%,Vimentin表达阳性率由(0.61±0.24)%增加到(28.53±3.52)%,经配对样本t检验,按a=0.05水准,差异均具有统计学意义(P均<0.001)。结论前列腺癌内分泌治疗控制肿瘤生长一段时间后,可以导致EMT特点的细胞数量增加,肿瘤侵袭转移能力增强,这可能是现有内分泌治疗最终失败的原因之一。

Abstract:Objective Imploring the change of the biomarkers of epithelial- mesenchymal transition (EMT) in human prostate cancer tissue after endocrine therapy. Methods 17 pairs of human prostate cancer tissue before and after endocrine therapy were collected in First Hospital of Shanxi Medical University from Jan 2006 to Dec 2012. Im-munohistochemistry SABC was used to detect the expression of EMT markers E Cadherin, N Cadherin and Vimentin in the cancer cells. Results After endocrine therapy, the number of E Cadherin positive cells decreased from (87.8± 5.4)% to (6.3±1.7)%.The number of N Cadherin positive cells increased from (1.2±0.6)% to (46.1±5.6)%. The number of Vimentin positive cells increased from (0.61±0.24)% to (28.53±3.52)%. The difference had significance (P〈0.001). Conclusion Endocrine therapy can control the growth of the tumor for a period, meanwhile the number the EMT positive cells increases, which promotes the invasion and metastasis of the tumor. This phenomenon could be one of the reasons of failure of existing endocrine therapy.
