天然植物药物IHA-01体外对微管聚合影响作用的研究

摘    要:目的研究天然植物药物IHA-01对猪脑微管蛋白的影响,探讨其可能的抗肿瘤机制。方法通过在体外对温度进行两次循环调控,应用超速离心机从新鲜猪脑中提取纯化出微管蛋白,体外建立微管蛋白亚单位聚合-解聚平衡系统,在此平衡系统中通过测量其吸光度值的变化来检测药物对微管的作用。结果 IHA-01抑制微管蛋白亚单位聚合过程中吸光光度值升高。结论抑制微管蛋白聚合可能是天然植物药物IHA-01抗癌机制之一。

Effect of the natural herb IHA-01 on tubulin depolymerization activity in vitro

Abstract:Objective To explore the effect of the natural herb IHA-01 on tubulin depolymerization activity in vitro.Methods The tubulin was extracted from the fresh pig brain tissue by ultracentrifuge after regulating the temperature two circulations in vitro.The polymerization-depolymerization balanced system of tubulin was established in vitro and the absorbance value of tubulin was measured to test the role of drug to tubulin.Results The IHA-01 could restrain the absorbance value of tubuin in polymerization process.Conclusion IHA-01 can inhibit the polymerization activity of tubulin.
