鼻咽癌组织中PD-1+淋巴细胞数升高的意义

摘    要:目的探讨鼻咽癌组织局部CD8+和PD-1+淋巴细胞在鼻咽癌发生发展中的作用。方法采用免疫组化方法检测64例鼻咽癌肿瘤组织和20例慢性鼻咽炎组织的CD8+和PD-1+淋巴细胞的分布情况。结果鼻咽癌肿瘤组织中CD8+淋巴细胞百分比为(10.68±6.81)%,与慢性鼻咽炎组织的(28.5±9.33)%比较,差异有统计学意义(P〈0.05);鼻咽癌组织中PD-1+淋巴细胞为(10.4±5.95)%,与鼻咽炎组织的(6.8±3.21)%比较,差异有统计学意义(P〈0.05)。鼻咽癌组织中PD-1+/CD8+淋巴细胞比率为(1.24±0.91),与鼻咽炎组织(0.28±0.2)比较,差异有统计学意义(P〈0.001),但PD-1+/CD8+淋巴细胞比率升高与鼻咽癌临床分期无关。结论鼻咽癌组织中CD8+淋巴细胞减少,PD-1+淋巴细胞增加,提示PD-1可能参与鼻咽癌免疫逃逸,与鼻咽癌的发生有关。

Abstract:Objective To investigate the expression and significance of PD-1 in nasopharyngeal carcinoma (NPC). Methods Immunohistochemistry method was used to detect CD8 and PD-1 expression of lymphocytes in 64 nasopharyngeal carcinomas and 20 chronic nasopharyngitis. Results Comparing with chronic nasopharyngitis, the lymphocytes with CD8 in NPC were decreased [(10.68±6.81)% vs (28.5±9.33)%,P〈0.05], while the lymphocytes with PD-1 were increased in NPC specimens [(6.8±3.21)% vs (10.4±5.95)%,P〈0.05]. The ratio of PD-1+/CD8+ lymphocytes was significantly up-regulated [(0.28±0.2) vs (1.24±0.91), P0.001] in NPC,but it was not significantly correlated with clinical stage. Conclusion This result suggests that PD-1 may play a role in immune evasion of nasopharyngeal carcinoma.
