针刺后处理对心肌缺血再灌注大鼠心脏组织腺苷A1受体的触发作用研究

目的:观察针刺对大鼠心脏组织A1ADR mRNA及A1ADR蛋白的影响,探讨针刺后处理对腺苷A1受体的触发作用,为针刺后处理的运用提供新的科学依据和研究靶点。方法:将36只SD大鼠,随机分为空白组、模型组、再灌注前针刺组、再灌注即刻针刺组、再灌注后针刺组、非经非穴针刺组,6只/组。采用荧光定量PCR技术检测大鼠心脏组织A1ADR mRNA的表达量,采用Western blot技术检测A1ADR蛋白的表达量。结果:1.A1ADR mRNA:再灌注前针刺与再灌注即刻针刺能明显上调A1ADR mRNA的表达量,且再灌注即刻针刺的上调最为明显(P0.05)。2.A1ADR蛋白:再灌注即刻针刺与再灌注后针刺能明显上调A1ADR蛋白的表达量,且再灌注即刻针刺的上调最为明显(P0.05)。结论:针刺后处理对腺苷A1受体有明显的触发作用,且以再灌注即刻针刺最为明显。

Study on Triggering Effect of Acupuncture on Adenosine A1 Receptor in Rats with Myocardial Ischemia-Reperfusion Injury

Objective:To observe the effect of acupuncture on adenosine A1 receptor in rats,and to explore the triggering effect of acupuncture posotonidiotning on the receptors,and to provide a new scientific basis for the application of acupuncture posotonidiotning.Methods:Thirty-six SD rats were randomly divided into blank group,model group,acupuncture before reperfusion group,acupuncture at reperfusion group,acupuncture after reperfusion group,non-acupoint group.The expression of A1ADR mRNA in rat heart were detected by RTFQ PCR.The expression of A1ADR protein was detected by Western blot.Results:1.A1ADR mRNA.The acupuncture befor reperfusion group and the acupuncture at reperfusion group could increase the expression of A1ADR mRNA,and the acupuncture at reperfusion group was the most obvious group (P ＜ 0.05).2.A1ADR protein.The acupuncture at reperfusion group and the acupuncture after reperfusion group could increase the expression of A1ADR protein,and the acupuncture at reperfusion group was the most obvious group (P ＜ 0.05).Conclusion:Acupuncture has obvious triggering effect on adenosine A1 receptor,and acupuncture at the reperfusion has the most obvious triggering effect.
