BMSC-肝细胞共培养上清治疗大鼠急性肝衰竭疗效观察

摘    要:目的探讨骨髓间充质干细胞(bonemarrow mesenchymal stemcells,BMSC)-肝细胞共培养上清治疗大鼠急性肝衰竭(acute liver failure,ALF)的疗效,为其临床应用提供实验依据。方法分别利用肝细胞、BMSC、BMSC-肝细胞培养上清治疗腹腔注射D-氨基半乳糖胺(D-gal)诱导的大鼠ALF模型,观察各治疗组动物的l周存活率,肝功能和肝组织病理改变。结果肝细胞、BMSC及两者共培养上清治疗组大鼠存活率显著高于对照组(P〈0.01),其中共培养上清治疗组大鼠存活率最高达80%;各治疗组ALT、AST、TBIL较治疗前均显著降低(P〈0.01),共培养上清治疗组下降最明显;各治疗组肝脏病理组织学损害明显减轻,对照组损害加重。培论BMSC-肝细胞共培养上清能更有效的救治大鼠ALF模型,有望为ALF的治疗提供了新的治疗途径。

Abstract:Objective To investigate the therapeutic potential of supernatant, which is derived from co - culture of bone marrow mesenchymal stem cells (BMSC) and hepatocytes, in the treatment of acute liver failure (ALF) rats. Methods Supernatants derived from hepatocytes, BMSC or co - culture of both were intraperitoneal injected into D -galaetosamine amine (D -gal) induced rats with the ALF phenotype. Survival rate, liver functions and histopathology were examined before or after transplantation. Results Supernatants from different cells can improve survival rate, liver functions and histology. Supernatant derived from co - culture of BMSC and hepatocytes was found to be more efficient than supernatants derived from BMSC or hepatocytes in the treatment of ALF (P 〈0.01 ). Conclu- sion Our finding suggest that supernatant injection of co - culture of BMSC and hepatocytes is a novel approach for treating acute liver failure and may be a promising therapeutic intervention in the future.
