盐酸小檗碱对HL-60细胞的诱导分化及其作用机制

摘    要:目的:研究盐酸小檗碱对人早幼粒白血病HL-60细胞诱导分化的作用及其分子机制。方法:HL-60细胞经盐酸小檗碱处理后,观察细胞形态学变化,以及细胞NBT还原能力的改变,用流式细胞仪测定细胞增殖周期、CD11b、C-myc、C-fos表达。结果:盐酸小檗碱作用于HL-60细胞后,形态学观察显示分化细胞的特征;NBT还原能力增强;CD11b表达升高;细胞被阻滞于G0/G1期,S期细胞数明显减少;C-myc基因表达减弱,C-fos基因表达增强。结论:盐酸小檗碱可诱导HL-60细胞分化成熟,其机制可能是通过调控与HL-60细胞增殖、分化相关基因表达,抑制DNA合成,从而抑制细胞增殖,诱导细胞分化。

Abstract:Aim:To investigate the induction of HL-60 cells differentiated by berberine hydrochloride and the possible molecular mechanism.Methods:After the treatment of HL-60 cells with berberine hydrochloride,the cell differentiation was observed for their cytomorphological changes and NBT reduction.Cell cycle,CD11b antigens and gene expressions(C-myc,C-fos)were analysed by the flow cytometry.Results:Berberine hydrochloride was found to induce differentiation of HL-60 cells.Morphological evaluation indicated that berberine hydrochloride treated cells exhibited granulation appearance.Both the positive rate of NBT reduction and the expression of CD11b increased.Flow cytometry analysis showed that berberine hydrochloride arrested HL60 cells in G_(0)/G_(1) phase,down-regulated C-myc gene expression and up-regulated C-fos gene expression.Conclusion:Berberine hydrochloride could induce the differentiation of HL-60 cells.Its molecular mechanism might be related to the modulation of gene expressions associated with the proliferation and differentiation,which leads to the inhibition of DNA synthesis.
