小鼠内源性抗原提呈树突状细胞疫苗的抗瘤作用

摘    要:目的:将pcDNA3-hCEA真核表达质粒和肿瘤来源的RNA分别转染小鼠骨髓树突状细胞(dendritic cell,DC),观察内源性抗原提DC疫苗诱导BALB/C小鼠对CT26-hCEA^ 的抗肿瘤免疫效应。方法:采用细胞因子rmGM-CSF和rmIL-4诱导培养法制备小鼠骨髓DC,利用Lipofectamine将pcDNA3-hCEA和肿瘤来源的RNA分别转染DC,制备能够内源性表达CEA的DC疫苗。使用DC疫苗预防免疫BALA/C小鼠,观察其对于荷CT26-hCEA^ 小鼠的抗肿瘤效应。结果:内源性抗原提呈DC疫苗明显延长荷CT26-hCEA^ 瘤小鼠生存期;与其他方法制备的DC疫苗相比,有较强的针对性抗肿瘤免疫效应和较长的持续效应。结论:地于免疫逃逸的肿瘤,内源性抗原提呈DC疫苗更具有免疫预防效应。

Abstract:Aim: To prepare the vaccine of dendritic cell (DC) transfected with pcDNA3 hCEA or RNA(CT26 hCEA +), and to observe the immune effects of the DC vaccine against CT26 hCEA + inoculated in BALB/C mice. Methods: DC from bone marrow was cultured in the presence of rmGM CSF and rmIL 4, and transfected with pcDNA3 hCEA or RNA(CT26 hCEA +). Vaccination with the DC, the survival time of the BALB/C mice with CT26 hCEA + was observed,and was compared to the mice with other DC vaccines. Results: The survival time of the mice vaccinated with the endogenous CEA presenting DC and CT26 hCEA + was prolonged by 40 d and the antitumor effects were more potent compared to other DC vaccines. Conclusions: The results demonstrate that the specific antitumor immune response of endogenous CEA presenting DC could be induced efficiently in the tumor evading immune reactivity.
