甲状腺素联合参桂甲减丸对甲状腺功能减退大鼠额叶内胆碱含量及突触蛋白损伤影响

摘    要:目的:分析甲状腺素联合参桂甲减丸对甲状腺功能减退大鼠额叶内胆碱含量及突触蛋白损伤影响。方法:选取SPF级Wistar雄性大鼠40只,随机数字表法将大鼠分成4组,正常组、模型组、甲状腺素组及联合用药组,每组各10只。除正常组外,其他各组大鼠制备甲减模型,第5周开始,甲状腺素组大鼠腹腔注射剂量为6g/100g的左旋甲状腺素(L-T4),每天1次,联合用药组在甲状腺素组基础上每天灌胃剂量为1ml/100g参桂甲减丸药液,模型组和正常组每天灌胃等剂量生理盐水,连续治疗2周。ELISA法检测血清促肾上腺素(ACTH)、皮质酮(CROT)、三碘甲状腺原氨酸(T3)、促甲状腺激素(TSH)、四碘甲状腺素原氨酸(T4)及促甲状腺激素释放激素(TRH)含量,碱性羟胺比色法检测额叶中乙酰胆碱含量,免疫组化检测额叶内munc-18表达状况。结果:和正常组相比,模型组大鼠血清TSH、TRH、CROT及ACTH含量上升,T3、T4含量下降;和模型组相比,甲状腺素组和联合用药组大鼠血清TSH、TRH、CROT及ACTH含量下降,T3、T4含量上升,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠乙酰胆碱含量下降;和模型组相比,联合用药组大鼠乙酰胆碱含量上升,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠Ⅱ、Ⅲ、Ⅳ及Ⅴ层额叶中munc-18的AOD数值下降;和模型组相比,联合用药组大鼠Ⅱ、Ⅲ、Ⅳ及Ⅴ层额叶中munc-18的AOD数值上升,差异均有统计学意义(P<0.05)。结论:参桂甲减丸结合甲状腺激素可恢复甲减大鼠额叶内受损突触蛋白,提升乙酰胆碱含量。

Abstract:Objective:To analyze the effect of Thyroxine combined with Shengui Jiajian Pill on choline content and synaptic protein damage in frontal lobe of hypothyroidism rats.Methods:Forty Wistar male rats of SPF grade were randomly divided into four groups:normal group,model group,thyroxine group and combination group,with 10 rats in each group.In addition to the normal group,the rats in the other groups were prepared for hypothyroidism.At the 5 th week,the thyroxine group was intraperitoneally injected with a dose of 6 g/100 g of levothyroxine(L-T4)once daily,and the combination group was in the thyroid gland.On the basis of the group,the daily dose was 1 ml/100 g Shengui Jiajian Pills,and the model group and the normal group were given normal saline daily for 2 weeks.Determination of serum adrenaline(ACTH),corticosterone(CROT),triiodothyronine(T3),thyroid stimulating hormone(TSH),tetraiodothyronine(T4)and thyroid stimulating hormone releasing hormone by ELISA(TRH)content,alkaline hydroxylamine colorimetric assay for acetylcholine content in frontal lobe,immunohistochemistry to detect munc-18 expression in frontal lobe.Results:Compared with the normal group,serum TSH,TRH,CROT and ACTH levels in the model group increased,and T3 and T4 levels decreased.Compared with the model group,serum TSH,TRH and CROT in the thyroxine group and the combination group.The levels of TTH and T4 decreased(P<0.05).Compared with the normal group,the acetylcholine content in the model group decreased.Compared with the model group,the acetylcholine content in the combination group was higher(P<0.05).Compared with the normal group,the AOD value of munc-18 in the frontal lobe of rats II,III,IV and V decreased in the model group;compared with the model group,the combination The AOD values of munc-18 in the frontal lobe of rats II,III,IV and V in the treatment group increased(P<0.05).Conclusion:Shengui Jiajian Pill combined with thyroid hormone can restore damaged synaptic protein in the frontal lobe of rats with hypothyroidism and increase the content of acetylcholine.
