自拟黄芪通络强心汤对冠心病血瘀证再灌注损伤模型大鼠实验研究

摘    要:目的:分析自拟黄芪通络强心汤对冠心病血瘀证再灌注损伤模型大鼠心肌保护作用及相关机制。方法:选取SPF级Wistar雄性大鼠60只,随机数字表法将大鼠分成4组,分别是模型组、正常组、阳性对照组与自拟黄芪通络强心汤组,每组各15只,除正常组外,其他3组大鼠制备心肌缺血血瘀证再灌注损伤模型,成功造模后阳性对照组大鼠灌胃0.10g/kg的复发丹参滴丸,自拟黄芪通络强心汤组灌胃6.8g/kg的自拟黄芪通络强心汤药液,模型组和正常组大鼠灌胃等剂量生理盐水,1次/d,持续给药14d。观察各组大鼠心律失常出现情况,检测心脏组织缝隙连接蛋白(Cx43)、血管紧张素Ⅱ1型受体(AT-1R)及血管紧张素Ⅱ(AngⅡ) mRNA相对表达量和心肌组织Cx43、AT-1R蛋白表达状况。结果:和模型组相比,阳性对照组、自拟黄芪通络强心汤组大鼠心律率降低,差异有统计学意义(P<0.05);和正常组相比,模型组大鼠心脏组织内AT-1R、AngⅡmRNA相对表达量升高,Cx43mRNA相对表达量降低;和模型组相比,阳性对照组、自拟黄芪通络强心汤组大鼠心脏组织内AT-1R、AngⅡmRNA相对表达量降低,Cx43mRNA相对表达量升高,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠心肌组织内Cx43蛋白表达升高,AT-1R蛋白表达降低;和模型组相比,阳性对照组、自拟黄芪通络强心汤组大鼠心肌组织内Cx43蛋白表达降低,AT-1R蛋白表达升高,差异有统计学意义(P<0.05)。结论:自拟黄芪通络强心汤可经过提高大鼠心肌细胞内Cx43含量,降低AT-1R、AngⅡ含量,降低心律失常发生率,起到保护心肌作用。

Experimental Study of Self-made Huangqi Tongluo Qiangxin Decoction for Model Rat of the Re-perfusion Injury of the Blood Stasis Syndrome of the Coronary Heart Disease

Abstract:Objective: To analyze the protective effect and mechanism of Self-made Huangqi Tongluo Qiangxin Decoction on myocardial protection in rats with coronary heart disease and blood stasis syndrome reperfusion injury. Methods: Sixty Wistar male rats of SPF grade were randomly divided into four groups: model group,normal group,positive controlled group and Self-made Huangqi Tongluo Qiangxin Decoction group. Each group consisted of 15rats. In addition to the normal group,the other three groups of rats were prepared for myocardial ischemia and blood stasis syndrome reperfusion injury model. After successful modeling, the rats in the positive controlled group were intragastrically administered with 0. 10g /kg recurrent Danshen Dripping Pills. The Self-made Huangqi Tongluo Qiangxin Decoction group was given 6. 8g /kg of Self-made Huangqi Tongluo Qiangxin Decoction. The rats in the model group and the normal group were given normal saline once a day for 14days. The arrhythmia of rats in each group was observed. The relative expression of gap junction protein ( Cx43),angiotensin II type 1receptor ( AT-1R) and angiotensin II ( AngII) mRNA and myocardial tissue Cx43,AT-1R protein expression status were detected. Results: Compared with the model group,the heart rate of the rats in the positive controlled group and the Self-made Huangqi Tongluo Qiangxin Decoction group decreased,the difference was statistically significant ( P<0. 05). Compared with the normal group,the heart tissue of the model group. The relative expression of AT-1R and AngII mRNA was increased,and the relative expression of Cx43mRNA was decreased. Compared with the model group,AT-1R and AngII mRNA in the heart tissue of the controlled group and the Self -made Huangqi Tongluo Qiangxin Decoction group. The relative expression of Cx43mRNA was increased,and the difference was statistically significant ( P<0. 05). Compared with the normal group,the expression of Cx43protein and the expression of AT-1R protein were decreased in the myocardial tissue of the model group. Compared with the model group,the expression of Cx43protein in the myocardial tissue of the controlled group and the Self-made Huangqi Tongluo Qiangxin Decoction group decreased, and the expression of AT- 1R protein increased,the difference was statistically significant ( P< 0. 05). Conclusion: Self-made Huangqi Tongluo Qiangxin Decoction can improve the content of Cx43in rat cardiomyocytes,decrease the content of AT-1R and AngII,reduce the incidence of arrhythmia,and protect the myocardium.
