薏仁降浊汤对急性痛风性关节炎大鼠关节组织内炎症因子影响

目的:分析薏仁降浊汤结合秋水仙碱和非布司他对急性痛风性关节炎大鼠关节组织内炎症因子影响。方法:选取由本院实验动物中心提供的SPF级Wistar雄性大鼠30只,制备急性痛风性关节炎模型,随机数字表法分成三组,对照组(10只)、西药组(10只)及联合组(10只),其中西药组大鼠每天灌胃1mg/kg非布司他、0.2mg/kg秋水仙碱,联合组大鼠在西药组基础上加用4.8g/kg薏仁降浊汤,对照组大鼠则每天灌胃等剂量生理盐水,连续灌胃7天。观察给药1、7天时大鼠急性炎症状况,全自动生化分析仪检测血清中性粒细胞、淋巴细胞及白细胞含量,ELISA法检测坏关节组织内TNF-α、IL-1β、NO含量及血清内NO、尿酸含量。结果:给药7天后西药组和联合组大鼠炎症指数、足趾肿胀度均低于对照组,差异有统计学意义(P<0.05),且联合组效果更优,和西药组对比差异有统计学意义(P<0.05);给药7天后联合组大鼠血清中性粒细胞、白细胞、尿酸及淋巴细胞含量较对照组、西药组显著降低,NO含量较对照组、西药组显著升高,差异均有统计学意义(P<0.05);给药7天后联合组大鼠踝关节组织内NO、IL-1β及TNF-α含量较对照组、西药组显著降低,差异均有统计学意义(P<0.05)。结论:薏仁降浊汤结合秋水仙碱和非布司他可显著降低急性痛风性大鼠关节组织内炎症因子含量。

Influence of Combination of Yire Jiangzhuo Decoction on Inflammatory Factors in Rats with Acute Gouty Arthritis

Objective: To analyze the effect of Yiren Jiangzhuo Decoction combined with Colchicine and Febuxostat on inflammatory factors in joint tissues of rats with acute gouty arthritis. Methods: Thirty male Wistar rats of SPF grade provided by the Experimental Animal Center of the hospital were selected to prepare an acute gouty arthritis model. The random number table method was divided into three groups: the controlle group(10 rats),the western medicine group(10 rats) and the joint group(10 rats),in which the western medicine group rats were fed with 1 mg/kg Febuxostat and 0.2 mg/kg Colchicine per day,and the rats in the combined group were treated with 4.8 g/kg Yiren Jiangzhuo Decoction on the basis of the western medicine group. Rats were given a daily dose of normal saline for 7 days. Observing the acute inflammatory conditions of rats at the 1 st and 7 th day of administration,detected the levels of serum neutrophils,lymphocytes and white blood cells by automatic biochemical analyzer,and detected the content and serum levels of TNF-α,IL-1β,NO in bad joint tissues by ELISA,and also internal NO,uric acid content. Results: After 7 days of administration,the inflammation index and toe swelling degree of the western medicine group and the combination group were lower than those of the controlled group,the difference was statistically significant(P<0.05),and the combined group had better results,and there was a difference between the western medicine group and the western medicine group. Statistical significance(P<0.05);Serum neutrophils,white blood cells,uric acid,and lymphocyte levels in the combination group were significantly lower than those in the controlled group and the western medicine group after 7 days of administration,and NO levels were significantly higher than those in the controlled group and the western medicine group. The difference was statistically significant(P<0.05). The contents of NO,IL-1β and TNF-α in the joints of the joint group were significantly lower than those of the controlled group and the western medicine group after 7 days of administration,and the differences were statistically significant(P<0.05). Conclusion: In combination with Colchicine and Febuxostat,Yiren Jiangzhuo Decoction can significantly reduce the content of inflammatory cytokines in the joints of rats with acute gouty.
