真武汤加味对糖尿病肾病大鼠肾组织中IGF-1表达的影响

摘    要:目的:探究真武汤加味对糖尿病肾病(DN)大鼠肾组织中胰岛素样生长因子-1(IGF-1)表达的影响。方法:腹腔注射链脲佐菌素建立DN大鼠模型,随机分为对照组、DN模型组、低剂量药物治疗组、高剂量药物治疗组。检测各组大鼠24小时尿蛋白总量(TP)、血糖(GLU)、血尿素氮(BUN)、血肌酐(SCr)等生化指标,采用Western Blot及荧光定量法测定各组大鼠肾组织IGF-1及其受体IGF-1R的表达量。结果:模型组、低剂量组及高剂量组大鼠的TP、BUN、SCr、GLU水平均显著高于阴性对照组(P<0.05);低剂量组及高剂量组各指标显著低于模型组(P<0.05);高剂量组各指标显著低于低剂量组(P<0.05);各组大鼠肾组织IGF-1、IGF-1R表达水平差异显著(P<0.05),各组大鼠IGF-1、IGF-1R表达量从高到低依次是模型组、低剂量组、高剂量组、阴性对照组。结论:真武汤加味能改善糖尿病肾病大鼠肾功能,其作用机制可能是通过抑制IGF-1、IGF-1R的过表达,起到对肾脏的保护作用,延缓DN的发展。

Abstract:Objective To explore the effect of modified Zhenwu Decoction on the expression of insulin-like growth factor-1(IGF-1) in renal tissues of rats with diabetic nephropathy(DN).Methods DN rat models were established by intraperitoneal injection of streptozotocin,and they were randomly divided into the control group,the model group,the low-dose group and the high-dose group.Biochemical indexes such as 24 h urinary total protein(TP),blood glucose(GLU),blood urea nitrogen(BUN) and serum creatinine(SCr) in all groups were detected.The expression levels of IGF-1 and its receptor IGF-1 R in renal tissues of rats in each group were detected by Western Blot and fluorescence quantitative method.Results Levels of TP,BUN,SCr and GLU were significantly higher in the model group,the low-dose group and the high-dose group than the negative control group(P<0.05),showing the high-dose grouP< the low-dose grouP< the model group(P<0.05).There were significant difference in expression levels of IGF-1 and IGF-1 R in renal tissues between groups(P<0.05),showing the model grouP> the low-dose grouP> the high-dose grouP> the negative control group.Conclusion Zhenwu Decoction can improve the renal function in rats with DN and the mechanism may be protecting the kidney and delaying the development of DN through inhibiting the overexpression of IGF-1 and IGF-1 R.
