摘    要:目的: 了解肺炎链球菌(S.pn)在宿主体内转化发生的情况,以便于深入研究体内细菌转化对其毒力的影响. 方法: 通过基因重组和转化构建绿色荧光报告细菌转化发生的S.pn菌株22G;通过酶切、转化实验、RT-PCR、测序等多种方法鉴定菌株构建成功;通过荧光显微镜观察荧光. 结果: 荧光蛋白可以报告细菌转化的发生;体内实验表明,细菌在宿主体内能够发生转化. 结论: 利用荧光报告细菌转化发生可以用于观察细菌体内转化发生的研究.

Abstract:AIM: To study when and where streptococcus pneumoniae (S.pn) forms transformation in vivo so as to study the influence of pneumococcal transformation on its virulence. METHODS: S.pn 22G whose transformation forming could be reported by enhanced green fluorescence protein (EGFP) was constructed and was identified by restricted enzyme digestion, transformation experiment, RT-PCR and DNA sequencing. Fluorescence was observed by fleorescence microscope. RESULTS: The fluorescence emitted by 22G which was isolated from BABL/c mice or cultured in vitro was observed. CONCLUSION: S.pn can form transformation in vivo and using EGFP as report protein, pneumococcal transformation forming can be detected in vivo.
