1 Integrin antisense oligonucleotides inhibit collagen gel contraction stimulated by AVP

AIM：To observe the influence of argipressin（AVP）on the collagen gel contraction by cardiac fibroblasts(CFs), and the interference effect of β1 integrin antisense oligonucleotides. METHODS：A three-dimensional culture system of cardiac fibroblasts from SD neonatal rats was set up, as well as phosphorothioate modified oligonucleotides and AVP were added to the collagen gel system, the index of contraction was calculated. RESULTS：①The contraction index was significantly increased by AVP (≥1×10-7mol/L, P<0.01 vs control); ②From the 4th hour AVP (1×10-7 mol/L) began to promote collagen gel contraction by cardiac fibroblasts (P<0.01, vs control) and continued at least 48 h; ③The index of contraction was significantly lower in the collagen gel treated with antisense oligonucleotides than that in the others. CONCLUSION：AVP can stimulate the collagen gel contraction of CFs, which was effectively inhibited by antisense oligonucleotides. These results indicated that β1 integrin plays a significant role in CFs migration promoted by AVP.
