摘    要:目的:介绍激光共聚焦显微镜技术及其在观察受体细胞分布实验中的应用。方法:采用原位杂交、免疫荧光双标结合激光共聚焦显微镜技术。结果:孤啡肽受体ORL1在成年大鼠的皮层,海马及下丘脑区域的神经元,星型胶质细胞,小胶质细胞上均有表达。结论:激光共聚焦显微镜技术及免疫荧光双标技术在检测受体的细胞分布方面是一种实用的、简单的方法。

Abstract:Objective\ To introduce laser confocal microscopy for detection of the distribution of opioid receptor\|like(ORL) receptors on neural cells.\ Methods\ The expression of ORL1 receptor mRNA in different neural cells was examined by the combination of in situ hybridization technique, immuno\|fluorescent double labeling and laser scanning confocal microscopy.\ Results\ ORL1 was found to express in cerebral cortex, hippocampus and hypothalamus of adult rats, particularly on astrocytes and microglial cells.\ Conclusion\ Confocal microscopy combined with immuno\|fluorescent double labeling techniques was a practical and preferred method to detect the distribution of receptors on the cells.
