茶氨酸对人宫颈癌Hela细胞增殖作用的影响

目的研究茶氨酸对人宫颈癌Hela细胞增殖和凋亡的影响。方法 MTT法测定茶氨酸不同浓度不同时间对Hela细胞增殖活性的影响;荧光显微镜观察Hela细胞的形态变化;流式细胞仪检测细胞凋亡率。结果茶氨酸对Hela细胞具有显著增殖抑制作用,并呈浓度和时间依赖性;荧光显微镜观察发现茶氨酸处理组的Hela细胞有典型的凋亡特征;流式细胞仪检测结果显示,与空白组比较,茶氨酸处理后的Hela细胞的凋亡率显著增加(P<0.01)。结论茶氨酸对人宫颈癌Hela细胞的增殖具有抑制作用,且其与诱导Hela细胞凋亡有关。

Effect of Theanine on the Proliferation of Human Cervical Carcinoma Hela Cells

Objective To study the effect of theanine on proliferation and apoptosis of cervical cancer Hela cells in vitro. Methods Cervical cancer Hela cells were cultured in vitro.MTT assay was used to investigate the inhibitory effect of theanine on cervical cancer Hela cells which were treated with different concentrations of theanine for 24,48 and 72 h.Hoechest 33342 fluorescent stainy was used to observe the morphological changes of Hela cells by Hoechest33342 flureseene ataning;and flow cytometry with AnnexinV-FITC/PI double staining was carried out to examine the apoptosis. Results Theanine(50 μmol/L,250 μmol/L,500 μmol/L) inhibited the proliferation of Hela cells in dose-dependent and time-dependent manner;In theanine-treated group(500 μmol/L),cell nucleus concentration and "apoptotiebody" could be seen by fluorescence microscope.Moreover,in theanine-treated group(500 μmol/L),compared with the control group,the apoptosis rate of Hela cells was significantly increased(P<0.01). Conclusion Theanine can significantly inhibit the proliferation of cervical cancer Hela cells and induce the apoptosis of cervical cancer Hela cells.
