模拟CO2气腹对人胃癌MKN-45细胞增殖和细胞周期的影响

目的 通过建立体外模拟CO2气腹环境，研究CO2气腹对人胃癌MKN-45细胞增殖和细胞周期的影响。方法 建立体外模拟CO2的气腹环境，在全自动气腹机作用下维持密闭培养箱内压力为12mmHg，作用时间4h。处理后用MTT法检测细胞增殖情况；通过流式细胞仪观察细胞周期变化。结果 MTT法检测细胞增殖结果显示，气腹组人胃癌MKN-45细胞的增殖速度与对照组比较无显著性差异（P〉0．05）；流式细胞仪检测人胃癌MKN-45细胞周期，CO2气腹组增殖指数与对照组比较无显著性差异（P〉0．05）。结论 体外模拟持续12mmHg CO2气腹环境，连续对人胃癌MNK-45细胞作用4h，不会促进胃癌细胞的增殖和生长。

Effect of mimic CO2 pneumoperitoneum on cell proliferation and cell cycle of human gastric cancer MKN-45 cells

Objective To study the effects of mimic carbon dioxide pneumoperitoneum on cell proliferation of human gastric cancer cells.Methods The gastric cancer cells MKN-45 were exposed to CO2 environment for 4 h under the pressure of 12 mmHg and those cultured in normal experiment condition as control.The cell proliferation of MKN-45 cells were examined by MTT method on day 1-7 after exposure and their cell cycle by flow cytometry 24 h after exposure.Results The cell proliferation and cell proliferative index(PI)showed no statistical difference between CO2 pneumoperitoneum group and control group(P>0.05).Conclusion The cell proliferation and cell cycle of human gastric cancer MKN-45 are not significantly affected after exposure to CO2 pneumoperitoneum.
