高效液相色谱法测定广西产匙羹藤果中牛弥菜醇A的含量

目的 建立匙羹藤果中牛弥菜醇A含量的测定方法,并对广西产匙羹藤果中的牛弥菜醇A含量进行测定.方法 采集广西7个产地的匙羹藤果进行检测.确定高效液相色谱法(HPLC)的测定条件,并采用正交实验优选牛弥菜醇A的提取方案.结果 以NH2柱(4.6 mm ×250 mm,5 μm)为色谱柱,乙腈-水(85 ∶15)为流动相,流速为1.0 ml·min-1,检测波长为207 nm,柱温为30℃,作为HPLC的测定条件.选择甲醇40%、超声时间提取60 min、溶剂体积60 ml作为提取方案.牛弥菜醇A在进样量为0.4004~20.02 μg之间呈良好的线性关系,平均回收率为100.43%.广西7个地产的匙羹藤果中中牛弥菜醇A的平均含量为34.24~56.57 mg/g,平均相对标准偏差为0.03%~2.37%.结论 所建立的含量测定方法可用于测定匙羹藤果中牛弥菜醇A的含量,为匙羹藤果质量标准的制定提供参考.

Quantitative determination of conduriol A in fruit of Gymnema sylvestre(Retz.) Schuit from Guangxi by high performance liquid chromatography

Objective To develop the quantitative determination method of conduritol A in the fruit of Gymnema sylvestre(Retz.)Schult,then to determine the content of conduritol A in the fruit of Gymnema sylvestre(Retz.) Schult from Guangxi.Methods The fruits of Gymnema sylvestre(Retz.) Schult were collected from 7 regions of Guangxi for determination.The determination condition of high performance liquid chromatography(HPLC) was decided,and the orthogonal experiment was used to optimize the extraction program of conduritol A.Results Chromatographic column of NH2 column(4.6 mm×250 mm,5 μm),mobile phase of acetonitrile-water(85 ∶15),flow rate of 1.0 ml/min,the detection wave of 207 nm and column temperature of 30 ℃ were decided as determination condition of HPLC.The concentration of formaldehyde,duration of ultrasonic extraction and solvent volume were set as 40%,60 minutes and 60 ml for extraction program respectively.A good linear relationship was found when the sample sizes of conduriol A were between 0.4004 μg and 20.02 μg,and the average recovery rate was 100.43%.Of Gymnema sylvestre(Retz.) Schult from 7 regions of Guangxi,the average contents of conduriol A in the fruits ranged from 34.24 to 56.57 mg/g,and the relative standard deviations were 0.03%-2.37%.Conclusion The quantitative determination method developed can be applied to determing the content of conduritol A in the fruit of Gymnema sylvestre(Retz.) Schult and provide the reference for formulating the quality standard of fruit of Gymnema sylvestre(Retz.) Schuit.
