摘    要:目的 探讨姜黄素对胃癌MGC-803细胞磷酸化-丝氨酸/苏氨酸蛋白激酶(p-AKT)表达的影响.方法 将MGC-803细胞分为空白对照组和四个药物实验组,各组姜黄素浓度分别为5 μmol/L,10 μmol/L,20 μmol/L,40 μmol/L.经不同浓度的姜黄素作用24 h、48 h、72 h后,应用CCK8法检测姜黄素对MGC-803细胞增殖的影响;应用Annexin V-FITC/PI双染色法测定不同浓度的姜黄素作用24 h后胃癌MGC-803细胞凋亡情况;应用蛋白免疫印迹法检测不同浓度姜黄素作用24 h对MGC-803细胞内p-AKT蛋白表达水平的影响.结果 姜黄素对MGC-803细胞增殖有抑制作用,且呈剂量依赖性和时间依赖性.在5~40 μmol/L浓度范围内,姜黄素呈浓度依赖性地促进MGC-803细胞的凋亡,并下调MGC-803细胞中p-AKT的水平.结论 姜黄素可能是通过抑制p-AKT蛋白的表达和AKT信号通路促进MGC-803细胞凋亡并抑制其增殖.

Abstract:Objective To investigate the impact of curcumin on the protein expression of phosphorylase serine threonine kinase,phosphorylated AKT(p-AKT),in gastric cancer MGC-803 cells.Methods The MGC-803 cells were divided into blank control group and four medicine experiment groups with various concentrations of curcumin(5,10,20 and 40 μmol/L,respectively).After the treatment with various concentrations of curcumin for 24,48 and 72 hours,cell counting kit 8(CCK8) assay was used to determine the effect of curcumin on the proliferation of MGC-803 cells;Annexin V-FITC/PI double staining to detect the apoptosis of MGC-803 cells after 24-hour treatment with various concentrations of curcumin;and Western blot to determine the effects of 24-hour treatment with various concentrations of curcumin on the expression levels of p-AKT protein in MGC-803 cells.Results Curcumin had an inhibitory effect on the proliferation of MGC-803 cells in a dose-and time-independent manner.Under the concentration range of 5 to 40 μmol/L,curcumin promoted the apoptosis of MGC-803 cells in a dose-dependent manner and down-regulated the level of p-AKT protein in MGC-803 cells.Conclusion Curcumin might play a role in promoting the apoptosis of MGC-803 cell and inhibiting its proliferation through suppressing the expression of p-AKT protein and AKT signaling pathway.
