摘    要:目的观察过热、过成饮食引起胃粘膜损伤以致造成胃粘膜萎缩的过程中超氧化物歧化酶(SOD)及丙二醛(MDA)的动态变化。方法采用55℃、15%NaCl连续灌胃12周,制成大鼠萎缩性胃炎模型,对照组采用等量的25。C水灌胃,在灌胃期间每隔4NJ处死一批大鼠。将大鼠麻醉后腹主动脉取血分离血清,同时摘取胃组织,制成组织匀浆,分别测定血清及胃黏膜组织SOD和MDA含量。SOD测定采用改良的盐酸羟胺法,总蛋白测定采用Lowry’s法测总蛋白,MDA按照试剂盒中的步骤严格操作。结果经热盐水灌饲大鼠的血清和胃黏膜组织中SOD活性和MDA均表现出了较好的时间效应关系,随处理时间延长,SOD活性显著降低和MDA含量明显升高。结论活性氧损伤在萎缩性胃炎发生发展过程中起着重要的先导作用。

The Dynamic Variation of SOD and MDA in Serum and Gastric Mueosa of Atrophic Gastritis Rat Caused by High-salt-hot Water

Abstract:Objective: To investigate the dynamic variation ofsuperoxide dismutase (SOD) and malondialdehyde (MDA) in the process of atrophic gastritis formation caused by over salt hot diet. Methods: The rat model of atrophic gastritis was made by feeding 55℃ 15% salty water for 12 weeks, the normal control group were made by feeding 25℃ water, and the rats in each group were killed group every 4 weeks. After the rat hocused, blood was taken form abdominal aorta and separated blood serum, meanwhile the stomach homogenate was done. The SOD and MDA in serum and gastric mucosa of atrophic gastritis rat were detected by improved muriatic acid hydroxylamine method and MDA kits, respectively. The total protein of stomach homogenate was tested by Lowry's method. Results: The SOD and MDA in serum and gastric mucosa had a perfect time-effect relation in hot-salt water group. The SOD and MDA decreased significantly in hot-salt water group in time-dependent manner. Conclusions: Active oxygen might play an important precursive effect in the formation of atrophic gastritis.
