异搏定逆转骨肉瘤多药耐药的体外初步观察

摘    要:目的:探讨异搏定是否能在体外逆转骨肉瘤多药耐药。方法:应用MTT法测定异搏定与阿霉素共同作用后骨肉瘤细胞的细胞活性。结果:1μg阿霉素单独作用24h,骨肉瘤细胞的细胞活性在90%左右,抑制率为10%,48h抑制率为50%左右,而加入异搏定后,骨肉瘤的细胞活性明显降低,特别是在1μg/ml细胞悬液浓度时,抑制率最高,24h细胞抑制率达50%左右。结论:异搏定体外可逆转骨肉瘤多药耐药。

Abstract:Objective: To determine whether verapamil (VER) can reverse the drug resistance in vitro.Metheds: The cytoactivity of human osteosarcoma cells incubated with doxorubicin (DOX) and verapamil together was detected by using the method of MTT colorimeritric analysis. Results:The cytoactivity of human osteosarcoma cells incubated with DOX for 24 hours was 90%, inhibiting rate (IR) being 1O%,and it was 50%for 48 hours,whereas when the cells were incubated with VER and DOX together,especially when the concentration of verapamil was 1 μg/ml,the IR for 24 hours was equal to that of 48 hours incubated with doxorubicinonly. Conclusion:Verapamil can reverse the drug resistance in cells of human osteosarcoma cell line (OS-732).
