慢性心理应激对实验大鼠行为和免疫功能的影响

目的探讨慢性心理应激对实验大鼠行为和免疫功能的影响。方法取健康雄性SD大鼠,随机分为两组:对照组(10只)、心理应激组(10只)以慢性束缚制动法建立慢性心理应激模型,通过旷场试验及悬尾试验观察各组大鼠在应激前后的行为变化;用放射免疫法测定血清皮质醇、白细胞介素-2、白细胞介素-6、白细胞介素-8的含量。结果(1)实验前两组大鼠体重无统计学意义;实验终止时,应激组大鼠体重显著低于对照组(P<0.05);(2)旷场实验结果显示,应激组大鼠的潜伏期明显延长(P<0.01),穿格数、直立次数、修饰时间显著减少(P<0.01),粪便粒数显著增加(P<0.05);(3)悬尾实验结果显示,应激组静止时间显著增长(P<0.01),挣扎次数显著减少(P<0.01);(4)应激组大鼠血清皮质醇含量显著高于对照组(P<0.01),而IL-2、IL-6、IL-8含量显著低于对照组(P<0.05)。结论慢性心理应激可引起大鼠体重增长缓慢、行为异常、释放过量皮质醇,使大鼠免疫功能受到抑制。

The Effect of Rat by Chronic Psychological Stress on Behavior and Immune Function

Objective To study the effect of chronic psychological stress on behavior and the cortisol,interleukin-2,interleukin-6 and interleukin-8 in rats.Methods SD rats was used to construct chronic psychological stress model.To observe the reactions by open-field test and the tail-suspension test and to detect the Contents of cortisol,IL-2,IL-6 and IL-8 of rats in two groups.Results(1)The plasma IL-2,IL-6 and IL-8 levels in chronic psychological stress group were significantly lower than that in control group(P<0.05),while their cortisol level was significantly higher than that in control group(P<0.01);(2)The results indicated that,there are significant differences between the stress model group and the control group by the open field test.The results of the tail suspension test were the same as the tail suspension test.Conclusion The rats weight of chronic psychological stress grown slowly and they also showed the abnormal behavior,the higher cortisol and their immune function was depressed.
