盐酸多奈哌齐对大鼠血管性痴呆的治疗作用

目的 探讨盐酸多奈哌齐对血管性痴呆（VD）是否有治疗作用。方法 结扎双侧颈总动脉16w，制作大鼠VD模型，通过Morris水迷宫实验、跳台实验及皮层和海马病理，观察盐酸多杂哌齐对VD的影响。结果 与模型组比，盐酸多奈哌齐组大鼠水迷宫第5及6天达到平台的潜伏期明显缩短（P〈0．05或P〈0．01），第2、3及6天达到平台的游程明显缩短（P〈0.05），第7天大鼠平台象限内的逗留时间明显增加（P〈0．05），寻找平台的策略由边缘型、随机型转为趋向型、直线型明显增快（P〈0．05或P〈0．01）；第1天、第2天跳台的错误次数明显减少（P〈0．05），第2天的错误潜伏期明显延长（P〈0．05）；盐酸多杂哌齐明显减轻VD大鼠皮层及海马的病理变化。结论 盐酸多奈哌齐对大鼠VD有一定的治疗作用。

Treating effect of Donepozil hydrochloride on vascular dementia rats

Objective To determine whether Donepezil hydrochloride have therapeutic effect on vascular dementia (VD) rats.Methods VD rats model was made by ligation of bilateral common carotid arteriae (CCA) for 16 weeks to observe the effect of Donepezil hydrochloride on VD by Morris water maze and Step-down test and pathology of rat cerebral cortex and hippocampus.Results Latency of Morris test shortened at the 5th day and 6th day (P<0.05,P<0.01),distance shortened at the 2nd day,3rd day and 6th day (P<0.05),staying time at the platform quadrant increased at the 7th day (P<0.05),seeking tactics improved from edge and random to trend and line type in the Morris test (P<0.05,P<0.01); latency prolonged at the 2nd day (P<0.05), error times decreased at the 1st and 2nd day in Step-down test (P<0.05) in Donepezil hydrochloride group; Donepezil hydrochloride reduced significantly pathological change in cerebral cortex and hippocampus of VD rats.Conclusions Donepezil hydrochloride has treating effect on VD rats.
