游离脂肪酸对胰岛BTC3细胞增殖及胰岛素分泌的影响

摘    要:目的:观察游离脂肪酸(FFAs)慢性作用对胰岛BTC3细胞增殖及胰岛素分泌的影响。方法:将不同浓度油酸作用于培养的胰岛BTC3细胞,用MTT法分析细胞增殖,流式细胞仪检测细胞周期,放免法检测基础和葡萄糖刺激的胰岛素分泌量(GSIS)。结果:与对照组相比,油酸作用后胰岛BTC3细胞MTF值明显低于对照组(P〈0.01),并且有剂量依赖性和时间依赖性。流式细胞仪分析显示油酸作用使BTC3细胞G1期细胞比例增加(P〈0.05),S期细胞比例明显减少(P〈0.01)。油酸作用后基础(2.8mmol/L葡萄糖)胰岛素分泌量明显增加,GSIS(16.7mmol/L葡萄糖)明显减少(均P〈0.01),并且均有剂量依赖性和时间依赖性。结论:油酸能够抑制胰岛BTC3细胞的增殖及GSIS,并且有剂量和时间依赖性。

Effect of free fatty acids on proliferation and insulin secretion of islet BTC3 cells

Abstract:Objective: To observe the chronic effect of high level free fatty acids (FFAs) on proliferation and insulin secretion of islet BTC3. Methods: Different concentration of oleic acid was administrated to the culture of BTC3 cells. MTT assay and flow cytometry (FCM) were used to detect cell proliferation and cell cycle. Radioimmunoassay was used to deter mine the amount of basal and glucose stimulated insulin secretion (GSIS). Results: Compared with the control group, the oleic acid groups significantly reduced the values of MTF absorbance in a time and dose-dependent manner (P〈0.01). FCM showed that the percentage of G1 phase cells increased (P〈0.05) and the percentage S phase cells markedly decreased (P〈0.01) after oleic acid treatment, the oleic acid group significantly increased basal insulin secretion (in the presence of 2.8 mmol/L) and markedly reduced glucose-stimulated insulin release (in the presence of 16.7 mmol/L) in a time and dose-dependent manner (all P〈0.01). Conclusion: Oleic acid can inhibit the proliferation of BTC3 cells and glucose-stimulated insulin release in a time and dose-dependent manner.
