银杏叶提取物对兔血管平滑肌细胞分泌细胞因子的影响

摘    要:目的 :探讨氧化低密度脂蛋白 (oxLDL)对兔血管平滑肌细胞 (VSMC)分泌细胞因子的影响 ,以及银杏叶提取物 (GbE)的干预作用。方法 :兔血管平滑肌细胞分 6组处理 ,以酶联免疫法检测培养上清液中细胞因子肿瘤坏死因子α(TNFα) ,白细胞介素 6(IL 6) ,白细胞介素 8(IL 8)水平。结果 :oxLDL(5 0mg·L-1)对TNFα,IL 6,IL 8分泌水平的增加于刺激后 8h达 6.4~ 8.5倍 ;GbE呈浓度 (1~ 2 0 0mg·L-1)、时间 (1~ 2 4h)依赖地抑制细胞因子分泌 ,并可显著降低oxLDL的上述作用 (与对照组比较均P <0 .0 5 )。结论 :oxLDL可促进兔VSMC分泌细胞因子TNFα,IL 6和IL 8。GbE可能通过抑制基础的及oxLDL刺激的细胞因子分泌对兔VSMC生物学功能发挥调控效用

Abstract:AIM: To investigate the effect of oxidized low density lipoprotein (oxLDL) on cytokins secretion of rabbit vascular smooth muscle cell (VSMC) and the intervention action of Ginkgobiloba extrat ( Gb E) METHODS: Rabbit aortic VSMC was cultured in six groups. The level of tumor necrosis factore alpha (TNF α), interleukin 6(IL 6) and interleukin 8(IL 8) were evaluated by ELISA method. RESULTS: oxLDL increased the secretion of TNFα, IL 6, and IL 8 by 6.4 8.5 fold in 8 h. Gb E decreased the cytockins secretion at a concentration of (1 200 mg·L -1 ) from 1 to 24 h in dependent manner and Gb E could inhibit the above effects of oxLDL. CONCLUSION:oxLDL can increase the secretion of TNF α, IL 6, and IL 8. Gb E may regulate the biology function of rabbit VSMC by inhibiting and cytokins secretion stimulated by oxLDL.
