| 摘 要： | 目的:研究肺癌组织中上皮钙粘蛋白(E-CD)基因启动子区5′CpG岛甲基化状况,揭示肺癌发生、发展过程中的分子水平变化和可能作用机制。方法:应用甲基化特异性PCR(methylation specific PCR)检测41例肺癌手术标本及同一病例正常肺组织中E-CD基因启动子区5′CpG岛甲基化水平。结果:E-CD基因启动子区甲基化阳性率在肺癌中显著高于正常肺组织,与肺癌的分型、细胞分化程度、淋巴结转移、TNM分期明显相关。结论:E-CD基因启动子区甲基化水平可作为较早期预测肺癌浸润转移程度,评价预后的有效指标。

| Abstract: | Objective: To study the methylation status of E-CD promoter region and discover the molecular changes and mechanism in lung cancer initiasion and progression.Methods: 41 lung cancer samples and matched normal lung tissue were detected by MSP.Results: The positive rate of CpG islands methylation status of E-CD gene promoter region in lung cancer was significantly higher than that in normal lung tissue,which was correlated with the lung cancer type,differentiation level,lymphonode metastasis status and TNM stage.Conclusion: The methylation status of E-CD gene promoter region is a good parameter for predicting invasion,metastasis and prognosis of lung cancer. |
