摘    要:目的分析低温液态条件下血浆对保存于葡萄糖甘露醇腺嘌呤(GMA)血细胞添加液中血小板体外质量的影响。方法取白膜法制备的浓缩血小板(PCs)7单位。每单位等分成3份,分离部分血浆后,按比例加入GMA血细胞添加液,制成10%血浆-PCs、35%血浆-PCs和100%血浆-PCs,置4℃冷藏5d。检测0d和5d的血小板计数、pH、平均血小板体积(MPV)、血小板低渗休克反应(HSR)、血小板形变能力(ESC)、P-选择素(P—selectin)、磷脂酰丝氨酸(PS)、血小板表面糖蛋白GPIbα以及乳酸和RANTES(Regulated On Activation,Normal,T—cell Expressed,and Secreted)的生成。结果4℃冷藏5d后,35%血浆-PCs与100%血浆-PCs比较,血小板计数、pH、MPV、HSR、P—selectin、GPIbα和PS以及乳酸和RANTES的差异无显著性(P〉0.05);而10%血浆-PCs与35%血浆-PCs和100%血浆-PCs比较,P—selectin和PS表达的增加,差异有显著性(P〈0.05),而HSR下降,但差异无显著性(P=0.09);10%血浆-PCs与100%血浆-PCs比较,血小板计数下降,乳酸的生成增加,差异有显著性(P〈0.05)。结论用GMA保存液4℃保存血小板时,35%ACD血浆可维持冷藏血小板的膜的完整性、正常代谢和功能。

Abstract:Objective To evaluate the effects of plasma in additive solution glucose mannitel adenine(GMA) on the in vitro quality of refrigerated platelet concentrates(PCs). Method Seven units of PLTs were prepared with bully coat methods. Each unit was split into three equal parts. After separating parts of plasma,GMA was added to achieve the 10% plasma-PCs, 35% plasma-PCs and 100% plasma-PCs, and stored at 4℃ for 5 days. On day 0 and day 5, PLT count, pH value, MPV, response to hypotonic shock (HSR), extent of shape chage (ESC), P-selectin and GPIba expression, phosphatidylserine (PS) exposure, lactate concentration and release of RANTES (regulated on activation, normal,T-cell expressed,and secreted)were determined. Result On day 5 stored at 4℃ ,no significant differences were seen in PLT count, pH value,MPV,HSR,P-selectin and GPIbα expression, PS exposure,release of lactate and RANTES between 35 % plasma-PCs and 100% plasma-PCs(P〉0. 05). Compared with 35% plasma-PCs and 100% plasma-PCs,P-selectin expression and PS exposure increased significantly in 10% plasma-PCs(P〈0.05),while HSR decreased but no significant difference occurrd. PLT count decreased and release of lactate increased in 10% plasma-PCs compared with 100% plasma-PCs(P〈 0.05). Conclusion Platelets stored in additive solution GMA with 35% plasma maintained their membrane integrity, metabolism and functions normally.
