川芎嗪通过减少肺癌A549细胞内活性氧的生成抑制细胞增殖及侵袭

摘    要:目的:探讨川芎嗪(tetramethylpyrazine,TMP)对人肺癌A549细胞生长侵袭的干预作用与活性氧(reactive oxygen species,ROS)及其调控蛋白表达水平的相关性。方法:采用CCK-8法检测川芎嗪对A549细胞生长的影响,采用Transwell小室实验检测川芎嗪对A549细胞侵袭能力的影响,采用DCFH-DA荧光探针检测川芎嗪对A549细胞内ROS水平的影响,Western-blot法检测川芎嗪对HIF-1α、VEGF-C、MMP-9蛋白表达水平的影响。结果:川芎嗪呈浓度依赖性抑制肺癌A549细胞增殖和侵袭能力(P<0.05),同时减少细胞内ROS生成(P<0.05),抑制HIF-1α、VEGF-C、MMP-9蛋白表达(P<0.05)。结论:川芎嗪可降低人肺癌A549细胞ROS水平,减轻氧化应激损伤,从而抑制肿瘤细胞的增殖及侵袭。

Abstract:Objective:To investigate the correlation between the intervention of TMP on the growth and invasion of A549 cells and the expression of reactive oxygen species(ROS) with regulated proteins level.Methods:To detect cell proliferation inhibition rate of A549 cells interfered with TMP in CCK-8.Transwell assay was used to determine the inhibitory effects of TMP on cell invasion.To observe intracellular ROS by fluorescence probe and expression of HIF-1α,VEGF-C,MMP-9 by Western-blot.Results:TMP inhibited A549 cells proliferation and invasion on dose(P<0.05).And the ROS production was also reduced with the expression of HIF-1α,VEGF-C,MMP-9 decreased(P<0.05).Conclusion:TMP can reduce the ROS level and oxidative stress injury of lung adenocarcinoma A549 cells,thus inhibit the invasion and proliferation of A549 cells.
