摘    要:目的:通过数据库挖掘来研究波形蛋白(Vimentin,VIM)基因在脑胶质瘤组织中的表达及其与临床预后的关系。方法:通过中国脑胶质瘤基因组图谱计划(Chinese Glioma Genome Atlas,CGGA)数据库(http://www.cgga.org.cn/)获取脑胶质瘤患者样本的mRNAseq数据及相应的临床数据,使用R(v.3.6.1)软件进行临床相关分析、生存分析、单因素和多因素Cox回归分析及ROC曲线评价VIM在胶质瘤进展中的预后作用,基因集富集分析VIM在脑胶质瘤中的功能,并行基因相关性分析。结果:分析发现VIM在脑胶质瘤组织中呈高表达,并与胶质瘤患者的不良预后相关;VIM可作为胶质瘤进展的独立预测因子。结论:VIM是预测脑胶质瘤患者预后的生物标志物,靶向胶质瘤VIM可能是抑制肿瘤进展的途径之一。

Abstract:Objective:To study the relationship between VIM gene expression and clinical prognosis in glioma tissues by database mining.Methods:The mRNAseq data and corresponding clinical data of glioma patient samples were obtained from the Chinese Glioma Genome Atlas(CGGA) database(http://www.cgga.org.cn/).R(v.3.6.1) software was used to analyze clinical correlation analysis,survival analysis,univariate and multivariate Cox regression analysis,and ROC curve to evaluate the prognostic role of VIM in glioma progression,and gene set enrichment analysis for VIM in glia tumor function,gene correlation analysis.Results:The analysis showed that VIM was highly expressed in glioma patients and was related to the poor prognosis of glioma patients.VIM can be used as an independent predictor of glioma progression.Conclusion:VIM is a biomarker for predicting the prognosis of glioma patients,and targeting glioma VIM may be one of the ways to inhibit tumor progression.
