AIM2在宫颈癌组织中的表达及其与HPV16病毒载量的相关性分析

摘    要:目的:研究AIM2与HPV16病毒载量在宫颈癌及其癌前病变组织中的关系。方法:通过免疫组化检测正常宫颈组织(50例)、LSIL(56例)、HSIL(108例)及宫颈癌组织(145例)中AIM2表达,采用实时荧光定量PCR法(硕世21 HPV 分型定量检测系统,BMRT)检测所有纳入患者HPV16型DNA病毒载量,分析AIM2蛋白表达水平、HPV16病毒载量在宫颈癌及其癌前病变中有无差异以及AIM2蛋白表达水平与HPV16病毒载量的相关性。结果:AIM2在正常宫颈组、LSIL组、HSIL组、宫颈癌组中的阳性表达率分别为12.0%(6/50)、26.8%(15/56)、82.4%(89/108)、84.1%(122/145),呈逐渐增高趋势,差异有统计学意义(χ2=207.675,P<0.001)。HPV16在正常宫颈组、LSIL组、HSIL组、宫颈癌组中的阳性表达率分别为16.0%(8/50)、30.4%(17/56)、44.4%(48/108)、47.6%(69/145),呈逐渐增高趋势,差异有统计学意义(χ2=18.567,P<0.001)。Spearman相关性分析可得:HPV16病毒载量与宫颈组织病变程度之间呈正相关(r=0.229,P<0.001),AIM2表达水平与HPV16病毒载量也呈正相关(r=0.467,P<0.001)。结论:宫颈病变程度与AIM2蛋白表达水平及HPV16病毒载量密切相关,AIM2蛋白表达水平及HPV16病毒载量随病变程度增加而增加。

Abstract:Objective:To investigate the relationship between AIM2 protein and HPV16 viral load in cervical cancer and its precancerous lesions.Methods:The immunohistochemistry was used to detect the expression of AIM2 in normal cervical tissues(50 cases),LSIL(56 cases),HSIL(108 cases) and cervical cancer tissues(145 cases).Real-time quantitative fluorescence PCR analysis(BioPerfectus Multiplex Real Time,BMRT) was used to detect the viral load of HPV16 in all patients.To analyze the difference of AIM2 protein expression level and HPV16 viral load in different cervical lesions and cervical cancer,and the correlation between AIM2 protein expression level and HPV16 viral load.Results:The positive expression rates of AIM2 in the normal cervix,LSIL,HSIL and cervical cancer groups was 12.0%(6/50),26.8%(15/56),82.4%(89/108) and 84.1%(122/145),respectively,with a trend of increasing gradually,which had and significant difference(χ2=207.675,P<0.001).The positive expression rates of HPV16 in the normal cervix,LSIL,HSIL and cervical cancer groups was 16.0%(8/50),30.4%(17/56),44.4%(48/108) and 47.6%(69/145),respectively,with a trend of increasing gradually,which had and significant difference(χ2=18.567,P<0.001).Spearman correlation analysis showed that there was a positive correlation between HPV16 viral load and cervical pathological changes(r=0.229,P<0.001),and there was a positive correlation between the expression level of AIM2 and HPV16 viral load(r=0.467,P<0.001).Conclusion:The degree of cervical lesion was closely related to the expression level of AIM2 protein and HPV16 viral load.The expression level of AIM2 protein and HPV16 viral load increased with the increase of cervical lesion degree.
