Abstract:The objective of this study was to investigate the effects of two Streptococcus suis type 2 strains, the virulent strain SS2-1 and the avirulent strain HA0609, on the intestinal microbiota in BALB/c mice. Three week old female mice were randomly divided into three groups: a control group (Mock), an SS2-1 strain infected group and an HA0609 strain infected group. The 16S rRNA v3-4 regions of the intestinal microbiota were sequenced on the Illumina Miseq platform, and the community structure and diversity of intestinal microbiota were analyzed according to the sequence data. The sequencing coverage of all samples in each group exceeded 99%, thus indicating that the experimental data were reliable. Marked differences were observed in the abundance and communities of intestinal microbiota among the three groups. Both the number of OTU and the alpha diversity of the intestinal microbiota were higher in the SS2-1 group than the control and HA0609 groups. The HA0609 group showed greater similarity to the control group. An increased proportion of pathogenic bacteria or potential pathogenic bacteria and a decreased proportion of probiotics were detected in the SS2-1 group, thus demonstrating the imbalance in the intestinal microbiota. According to the results of LEfSe analysis, Alphaproteobacteria may be used as a potential biomarker for diagnosis of Streptococcus suis type 2 infection. The results of this study not only provide insights into the abundance, diversity and composition of the intestinal microbiota of BALB/c mice infected with Streptococcus suis type 2 virulent or avirulent strains, but also identify the potential biomarker Alphaproteobacteria for diagnosis of Streptococcus suis type 2. This study provides experimental data for improving treatment and preventing Streptococcus suis type 2 infection by adjusting microorganisms.
