摘    要:目的建立玄参HPLC指纹图谱,以系统指纹定量法评价其质量。方法采用RP—HPLC法,以CenturySILC18BDS(200mm×4.6mm,5μm)色谱柱,1%乙酸-水和1%乙酸-乙腈为流动相,低压线性梯度洗脱,流速1.0mL·min^-1,柱温(30±0.15)℃,进样量5μL,紫外检测波长280nm。结果以哈巴俄苷为参照物峰,确定29个共有峰,建立了玄参HPLC指纹图谱。以10批玄参HPLC指纹图谱按平均值法生成的对照指纹图谱(RFP)为标准,用系统指纹定量法评价不同产地玄参质量,采用SPSS软件以Sm和Pm为指标进行系统聚类分析,10批玄参药材被分为四类。第1类含量偏低,第Ⅲ类含量偏高,第Ⅱ类质量最好,第Ⅳ类为不合格药材。结论系统指纹定量法以整体定性分析和整体定量分析密切结合,适用于玄参药材系统化学指纹的整体质量控制。

Abstract:Objective To establish HPLC fingerprints of Raidix Scrophulariae and to evaluate its quality by systematic quantified fingerprint method. Methods Chromatographic fingerprints were obtained by injecting 5 μL of the sample solution each time on a Century SIL C18 BDS column (200 mm×4.6 mm, 5 μm) with the gradient elution solvent system composing of 1% acetic acid water and 1% acetic acid acetonitrile. The flow rate was 1.0 mL . min^-1, the column temperature was maintained at (30±0. 15) ℃ and the detection wavelength was set at 280 nm. Results Twenty-nine co-possessing peaks were selected as the fingerprint peaks of Raidix Scrophulariae when harpagoside peak served as the referential peak to establish the HPLC fingerprints. The referential fingerprint (RFP) was synthesized by the mean values of 10 batches of Raidix Scrophulariae, which was taken as the qualified model to evaluate the quality of Raidix Scrophulariae from different places by systematically quantified fingerprint method. According to classification depending on the values of macro qualitative similarity (Sm) and macro quantitative similarity (Pm), 10 batches of Raidix Scrophulariae were divided into 4 categories. Conclusion Systematic quantified fingerprint method which is based on the close combination of macro qualitative analysis and macro quantitative analysis can effectively assess the quality of Raidix Scrophulariae.
