阿魏酸在正常和脾虚血淤大鼠体内的药动学及对胃肠激素的影响

摘    要: 目的 研究阿魏酸在不同“证”大鼠体内药动学的差异并初步探索其机制。方法 以高效液相色谱法分析阿魏酸在正常及脾虚血淤大鼠体内的血药浓度,同时以放免法检测3组大鼠血浆及肠道中胃动素和前列腺素E2含量的变化。结果 阿魏酸在脾虚血淤组大鼠体内的分布容积显著减小,消除速率显著降低,曲线下面积显著增高;多剂量静注阿魏酸后脾虚血淤大鼠血浆和近段空肠组织中胃动素的含量显著下降,降结肠和直肠组织中前列腺素E2的含量显著升高。结论 阿魏酸在正常及血淤证大鼠体内药动学存在差异,阿魏酸治疗血淤证作用与调正胃肠激素平衡有关。

Abstract:OBJECTIVE To study the pharmacokinetics of ferul ic Acid (FA) and its mechanism in rats with blood stasis due to hypofunction of th e spleen.METHODS The rats were divided into 3 groups:control group,blood stasis group and treatment group.The concentrations of FA in blood were analyzed by HPLC,while RIA measured the changing of gastrin and PGE 2 in p lasma and intestinal tissue.RESULTS The distribution volume and elimination of FA in the blood stasis group was significantly decreased,and the area under the curve was increased compared with the control group.After con tinuously intravascular injection FA,the gastrin in plasma and tissue of proxima l jejunum of blood stasis group was significantly decreased but PGE 2 in the ti ssue of descending colon and rectrum was increased compared with the control.CONCLUSION There was a distinct difference of the pharmacokinet ics of FA between the control group and the blood stasis group.The effect of FA on blood stasis due to hypofunction of the spleen was related to its regulation of balance of gastrointestinal hormones.
