自身免疫性肝炎患者血NK细胞频率及表型特征分析

目的探讨自身免疫性肝炎（AIH）中NK细胞在外周血中频率及表型特点。方法选择15例自身免疫性肝炎患者,采集入选对象外周抗凝血5ml,分离外周血单核细胞（PBMC）,进行CD56-PE、CD3-APC、CD16-FITC、HLA-DR-PERCP单克隆抗体4色染色,采用流式细胞技术进行检测。结果 AIH组与正常对照组NK细胞中CD56bright比例分别为19.42%±7.36%,6.06%±2.63%,AIH组显著高于正常对照组（P〈0.001）。AIH组与正常对照组,CD56bright细胞群中活化指标HLA-DR＋比例分别为6.13%±5.67%和2.10%±1.24%,AIH组显著高于对照组（P=0.017）。对CD56bright表达频率与自身免疫性肝炎患者肝功生化指标之间进行相关性分析,尚未发现两者之间的相关关系。结论 AIH患者外周血中CD56bright及其中活化细胞的比例均有增高,提示NK细胞可能通过活化的CD56bright分泌细胞因子途径在发病过程中发挥作用。

The frequency and phenotype of NK cells in peripheral blood of autoimmune hepatitis patients

Objective To discuss the character of frequency and phenotype of NK in the patients with autoimmune hepatitis.Methods Fifteen AIH patients from Beijing You＇an hospital from 2006 to 2008 were enrolled in this study.PBMC from AIH patients and healthy donors were detected for the phenotypes and frequency of NK by flow cytometric analysis with CD56-PE,CD3-APC,CD16-FITC,HLA-DR-PERCP four color staining.Results The proportion of CD56bright in NK cells from AIH patients and healthy control were 19.42%±7.36%,6.06%±2.63%,respectively and the difference between the AIH group and healthy control was significant （P 0.001）.The frequency of HLA-DR＋ in CD56bright cells in AIH patients was 6.13%±5.67%,significantly higher than that in healthy controls（2.10%±1.24%,P=0.017）.There was no relationship between the frequency of CD56bright cells and biochemical parameter in AIH patients.Conclusions These results suggest the increase in frequency of CD56bright and active proportion in CD56bright might play a role in the pathogenesis of AIH by secreting cytokines.
