摘    要:目的:验证microRNA-203在膀胱癌组织中的表达水平,并观察microRNA-203对膀胱癌细胞T24增殖和转移能力的影响。方法:选取膀胱癌组织和正常组织各20例,通过实时定量PCR分析microRNA-203的表达。通过转染上调microRNA-203在T24细胞中的表达,并通过PCR验证转染效率。再进一步通过CCK-8法检测microRNA-203上调后细胞增殖能力的变化。采用Annexin V-PI双染法观察microRNA-203对膀胱癌细胞凋亡的影响,采用transwell法观察microRNA-203对膀胱癌细胞转移能力的影响。结果:与正常组织相比,膀胱癌组织的microRNA-203表达明显降低(P0,.05)。上调T24细胞microRNA-203表达后,与对照组相比,细胞增殖能力明显减弱,并且可观察到有凋亡现象发生。进一步的transwell法可观察到microRNA-203可明显抑制T24细胞侵袭。结论:microRNA-203在膀胱癌的发生发展过程中发挥着明显的抑癌作用。

Abstract:Objective:To investigate the expression of microRNA-203 in bladder cancer tissues,and to detect the effect of microRNA-203 on the proliferation and invasion of T24 cell line.Method:The expression of microRNA-203 in nomal bladder and bladder cancer tissues were measured by qRT-PCR.Then,the effect of microRNA-203 on the proliferation of T24 was evaluated by CCK-8assay after transfection.The early apoptosis induced by PA-MSHA was measured by Annexin V-PI double staining,and Transwell assay was performed to detect the impact of microRNA-203 on the invasion.Result:The expression of microRNA-203 in bladder cancer tissus decreased.We found upregulation of microRNA-203 induced cell apoptosis and cell growth inhibition,and further reversed invasion in bladder cancer cell line.Conclusion:MicroRNA-203 plays an important role in the inhibition of bladder cancer growth.
