人牙周膜干细胞的分离培养及鉴定

目的:探讨人牙周膜干细胞的多向分化潜能。方法:体外分离培养人牙周膜细胞,光镜下及HE染色鉴定细胞形态学特征。免疫组化法检测波形蛋白、角蛋白、CD44的表达。成骨诱导法培养人牙周膜干细胞向成骨细胞分化。结果:体外成功分离培养人牙周膜干细胞。角蛋白阴性表达,波形蛋白阳性表达,CD44阳性表达。诱导成骨结果显示人牙周膜干细胞具有潜在的多向分化潜能。结论:人牙周膜干细胞在体外可以诱导分化为成骨样细胞,具有多向分化的潜能。

Isolation and verification of human periodontal ligament stem cells

Objective: To isolate and verification the human periodontal ligament stem cells(PDLSC)and investigate the differentiation potentials of PDLSC.Methods: PDLSC were isolated and cultivated in vitro.The cell morphology characteristics were observed under light microscope and by HE-stained.The expression of vimentin,ceratin and CD44 were detected by immunohistochemistry staining.The biologic activity of BMSCs was used by osteogenic differentiation and steatogenic differentiation.Results: PDLSC were isolated cultured successfully in vitro.Immunohistochemistry staining of vimentin were shown positive stain,ceratin were shown negative stain and CD44 were shown positive stain.Osteogenic differentiation shown osteogenic ability and positive stain of ALP.Conclusion: PDLSC can be induced into ossify-like cells in vitro.PDLSC have the capability of multilineage differentiations.
