摘    要:目的检测versican之多糖链在小鼠磨牙牙胚发育过程中的时空变化。方法取胎龄分别为E11.5,E12.0,E13.5,E15.0,E16.5和E18.5 d的ICR胎鼠头部,常规组织学处理,制备5μm厚矢状或冠状切片,用免疫组织化学方法检测versican核心蛋白与其硫酸软骨素多糖链在下颌第一磨牙牙胚组织中的时空表达。结果从小鼠下颌第一磨牙牙胚开始发生到钟状晚期,versican核心蛋白与4-硫酸软骨素在牙源性上皮及其衍生组织共表达。在牙乳头间充质细胞中versican核心蛋白开始与6-硫酸软骨素共表达,从钟状早期4-硫酸软骨素开始在内釉细胞深层的牙乳头间充质细胞间表达,且其与6-硫酸软骨素呈互补表达模式。结论 versican之硫酸软骨素多糖链在牙胚发生过程中硫酸化模式呈现时间-空间特异性改变,这种特异性改变可能与小鼠磨牙牙胚的形态发生以及细胞增殖与分化密切相关。

Abstract:Objective To investigate the temporal-spatial changes of the chondroitin sulfate side chains of versican in the development of embryonic first lower mouse molars. Methods Serial paraffin sections of 5 (xm of the first lower molars at different embryonic stages were made. The expression patterns of versican core protein, 4-chondroitin sulfate (C4S) and 6-chondroitin sulfate (C6S) were analyzed by immunohistochemistry. Results Versican core protein and C4S were colocalized in the dental epithelium and its derived tissues from the tooth initial stage to the late bell stage. While versican core protein and C6S were co-expressed in the dental mesenchyme tissues from bud stage to early bell stage, then C4S was able to be detected in the dental mesenchyme cells, which were adjacent to the inner enamel cells from bell stage. The expression patterns of C4S and C6S in the dental papilla were mutually complementary. Conclusion The expression patterns of the chondroitin sulfate side chains of versican in first lower mouse molars are temporal and spatial specific. This specific patterns may be involved in the tooth morphogenesis, cell proliferation and cell differentiation of the mouse molars.
