动脉粥样硬化大鼠“痰瘀”病理演变与相关基因表达研究

摘    要:目的:探讨动脉粥样硬化(atheroselerosis,AS)大鼠痰瘀病理演变规律和形成机制。方法:复制AS模型大鼠,通过光镜、电镜观察主动脉组织学改变;运用RT-PCR法检测相关基因表达。结果:随着AS的进展,血脂增高、血液流变学异常逐渐加重;在造模第17天脂质代谢明显异常,造模第22天血液流变学显著异常,第32天动脉硬化程度达到2级;基因c-myc、PDGF-A、c-fos、Bcl-2mRNA表达量依次上调,p53mRNA表达下调。结论:基因c-myc、PDGF-A、c-fos、Bcl-2mRNA表达上调、p53 mRNA表达下调可能是AS痰瘀病理演变的分子基础。

Abstract:Objective:To explore the molecular mechanisms of atherosclerosis (AS). Methods: AS rat model was made, arterial morphological histology changes were observed under optic and electronic microscope, and some genes were detected by RT-PCR. Results: There was biochemical change in the blood stasis-phlegm syndrome, which was associated with the alterations of tissue structure.
