摘    要:目的:从分子水平鉴别冬虫夏草与其混淆品北虫草。方法:从冬虫夏草及其常见混淆品北虫草中提取DNA,以核基因组通用引物为引物进行扩增,扩增产物经纯化后,用PCR产物直接测序法进行测序。结果:测得各样品rDNA的ITS及5.8S rDNA完全序列,18S和28S rDNA部分序列,分别为冬虫夏草459bp、北虫草548bp;二者ITS具显著差异,其中ITS1差异性达33.31%、ITS2差异性达26.67%。结论:ITS序列可有效地鉴别冬虫夏草及其混淆品北虫草。

Abstract:Objective To analyze the ITS sequence of Cordyceps and its adulterants Cordyceps mili-taris in order to provide molecular evidence for its identification.Methods Firstly,total DNA in the Cordyceps and Cordyceps militaris was extracted.Secondly,the ITS sequence was amplified by PCR with universal primer of ITS and sequenced after purification.Results The total length of ITS sequ-ence,Cordyceps was 459bp and adulterants was 548bp,such as 5.8S,18S,and 28S rDNA.Multiple s-equencing was alignment shows that there were much sequence variation between Cordyceps and its ad-ulterants.The variability of ITS1 was 33.31%,ITS2 was 26.67%.Conclusion ITS Sequence can be used to identify Cor-dyceps and Cordyceps militaris.
