戊四氮诱导单次惊厥幼鼠海马碱性成纤维细胞生长因子表达的研究

摘    要:目的利用戊四氮单次惊厥幼鼠模型,研究癫痫单次惊厥发作后海马各区碱性成纤维细胞生长因子(bFGF)表达情况。方法建立戊四氮腹腔注射幼鼠单次惊厥发作模型,分成A组(癫痫发作组)和B组(生理盐水对照组)。每个组后按造模后1天、7天、14天分为3组。HE染色观察病理形态改变,免疫组化观察海马bFGF表达变化。结果病理形态上A组可见海马神经元变性、坏死,胶质细胞增生,B组形态学无明显变化。bFGF表达:A组发作后1天bFGF表达增高最明显(P〈0.01),以CA1区和齿状回明显,以后逐渐下降,7天时仍高于B组(P〈0.05),14天时下降到较低水平,和B组无显著性差异(P〉0.05)。B组各时间点bFGF表达无显著性差异。结论幼鼠单次惊厥发作后海马bFGF表达增加,在发作后7天内较为明显。bFGF在癫痫后脑损伤的自我修复中可能发挥着重要的作用。

Abstract:Objective To study the expression of basic fibroblast growth factor(bFGF) in the hippocampus of epileptic pedo-rats at different time points and investigate its bearing on epilepsy.Methods Forty-two 14-day old SD pedo-rats were randomly divided into two groups: A(epilepsy model group) and B(normal saline group).Pentetrazole was used to induce seizure models.The expression of bFGF in the hippocampus of rats was studied by immunofluorescent labeling technique.H-E staining was done for pathomorphological study.Results In group A,neuronal degeneration and necrosis in the hippocampus,and cellular proliferation of glial cells as well,were noticed;the expression of bFGF was increased,most marked 1 day after seizure(P<0.01),continued to be higher for 7 days(P<0.05),and then declined and approached the control level 14 days later(P>0.05).Conclusion The expression of bFGF in hippocampus of 14-day rat is increased after acute epilepsy,significantly higher for 7 days after seizure.bFGF may play an important role in the self-plerosis after brain injury induced by epilepsy in pedo-rats.
