山东地区汉族2型糖尿病与GCK、FABP_2基因多态性的相关性研究

摘    要:运用聚合酶链反应 (PCR)技术 ,结合短串联重复序列 (STR)多态性标记的方法 ,对山东地区汉族 10 7例 2型糖尿病患者和 10 5例正常对照者 ,进行葡萄糖激酶 (GCK)基因及小肠脂肪酸结合蛋白 (FABP2 )基因的多态性分析。结果显示 GCK等位基因 A5与 2型糖尿病组呈正相关 (RR=15 .70 ,P<0 .0 0 1) ;GCK等位基因 A1、A3与 2型糖尿病组呈负相关 (RR=0 .2 8,P<0 .0 0 1;RR=0 .15 ,P<0 .0 0 1)。FABP2各等位基因频率在两组分布未见显著性差异。多因素非条件 L ogistic回归分析发现 ,2型糖尿病的发病与 GCK等位基因 A5呈显著正相关 ;与 GCK等位基因 A3呈显著负相关。年龄、甘油三酯、载脂蛋白 B及脂蛋白 a亦是 2型糖尿病发病的危险因素。认为 GCK基因多态性与山东地区汉族 2型糖尿病具有相关性 ,是 2型糖尿病的危险因素。FABP2 基因与 2型糖尿病未见相关性。

Abstract:Two STR region polymorphism were analyzed with the polymerase chain reaction and polyacrylamide sequencing gels in 107 subjects with type 2 diabetes mellitus and 105 nondiabetic subjects All subjects were biologically unrelated Chinese living Shandong without family history Results showed 5 alleles and 9 genotypes of GCK gene locus ,4 alleles 7 genotypes of FABP 2 gene locus were observed The differences in allelic frequencies of GCK gene between individuals with type 2 diabetes and nondiabetic individuals were compared(p<0 001) No difference was found in the frequency of FABP 2 alleles between the two groups(p<0 05) A5 allele of GCK gene was associated with type 2 diabetes (RR=15 70,p<0 001) Logistic regression analysis showed A5 allele was associated with type 2 diabetes We conclude ploymorphic microastellite repeat markers at the glucokinase gene locus is positively associated with type 2 diabetes in Chinese The polymorphism at the FABP 2 locus is not genetic marker for type 2 diabetes in Chinese
