实验性糖尿病肾病层粘连蛋白B1mRNA的变化及葛根素对其调节研究

摘    要:目的观察实验性糖尿病肾病鼠层粘连蛋白B1mRNA的变化及葛根素对其影响的研究。方法将实验用SD大鼠采用随机数字法分为正常对照组和造模组,造模组又分为糖尿病组和葛根素治疗组,治疗12周。观察治疗期间及治疗后大鼠的血糖、内生肌酐清除率、24h尿蛋白排泄率,原位杂交检测肾组织层粘连蛋白B.基因表达。结果造模组大鼠均出现肾脏功能损害;糖尿病组及葛根素治疗组尿白蛋白排泄率、内生肌酐清除率、层粘连蛋白B1mRNA、糖基化终未产物均与对照组比较有统计学意义(P〈0.05);葛根素治疗组较糖尿病组比较有统计学意义(P〈0.05)。结论葛根素通过抑制肾层粘连蛋白B1mRNA的表达对糖尿病大鼠肾脏起到保护作用。

Abstract:Objective To observe the effect of Puerarin on laminin B1mRNA in Streptozotoein-induced diabetic nephropathy rats.Methods Rats were randomly dirided into control and experimental groups.The latter was divided into two subgroups,namely diabetes and Puerarin groups.Both groups were treated for 12 weeks.We studied blood glucose levels,urinary albumin excretion rate within 24 hours.creatinine clearance rate,and expression of laminin B1 gene in kidney tissue determined by in situ hybridization analysis during and after the treatment.Results All rats in experimental groups exhibit renal dysfunction.Urinary albumin excretion rate,creatinine cleKrance rate,end products of lamininin B1 mRNA in diabetes and Puerarin groups were significantly different as compared with that in control groups.Conclusion Puerarin plays a certain role in the protection of kidneys in diabetic rats via inhibiting the expression of kidney lamininin B1.
