肺癌组织p53V、p16基因表达与DNA倍体的相关性及其临床意义

摘    要:目的:探讨肺癌组织细胞肿瘤相关基因p53V、p16基因的表达与DNA倍体类型的相关性及其临床意义。方法:采用流式细胞术对63例非小细胞肺癌患者瘤组织p53V 、p16 细胞表达率和DNA含量进行了检测,并分析了p53V、p16基因表达与肿瘤细胞DNA倍体类型、S期细胞比例(SPF)、凋亡指数(AI)及肿瘤是否转移的关系。结果:肺癌组织细胞的p53V 细胞检出率、DNA指数(DI)及SPF均显著高于正常对照组(P<0.01),肿瘤转移者也显著高于未转移者(P<0.01),而AI分别显著低于正常对照组及未转移者(P<0.01);随着DNA倍体从D→ND→T→AN→M,p53V 细胞检出率逐渐增高。肺癌患者瘤组织p16 细胞检出率的变化规律与p53V的变化恰好相反。二者相关性分析显示,p53V 细胞与p16 细胞表达率呈显著负相关(r=-0.712,P<0.05)。结论:肺癌患者瘤组织细胞p53V和p16基因编码蛋白的表达水平与肿瘤细胞生物学特性关系密切,不仅可用于肺癌发生的分子机制研究,而且还可以为肺癌恶性度、转移、预后判断提供可靠的客观依据。

Abstract:Objective:To explore the correlation between the expression of tumor related genes p53V and p16 and DNA contents in tumor tissues from patients with lung cancer and its significance. Methods:The expression rates of p53V and p16 cells and the DNA contents in tumor tissues of 63 patients with non-small cell lung cancer were measured and analyzed by flow cytometry. The correlation of the expression rates p53V and p16 cells with DNA contents, S-phase fraction(SPF),apoptotic index(AI) and metastasis were analyzed. Results:The expression rates of p53V ,DI and SPF in tumor tissues were higher than those of normal controls , and the expression rates in patients with metastasis were higher than those in patients without metastasis, respectively(P < 0.01),while the expression rate of AI was significantly lower than that of normal controls or patients without metastasis. The expression rate of p53V cells was highest and the expression rate of p16 cells was lowest in patients with the DNA content type of M, followed by AN, T, ND and D. The correlation analysis showed that the expression rate of p53V cells was negatively correlated to p16 cells( r = -0.712,P < 0.05). Conclusion:The expression of p53V and p16 genes coding protein in tumor tissues are so closely related to the biological characteristics of tumor cells of patients with lung cancer that the measurement of p53V and p16 expression may not only be used to study the molecular mechanism of lung cancer, but also provide important evidence for tumor malignant degree, metastasis and prognosis.
