BMP2、VEGF165双基因共表达质粒在小鼠骨髓基质干细胞的表达

摘    要:目的观察转染BMP2和VEGF165双基因共表达质粒在小鼠骨髓基质干细胞的表达情况。方法脂质体介导下将双基因真核表达质粒pIRES-BMP2-VEGF165导入小鼠骨髓基质干细胞,用RT-PCR和免疫组织化学方法观察BMP2和VEGF165双基因在小鼠骨髓基质干细胞内的表达。结果转染BMP2、VEGF165的小鼠骨髓基质干细胞有明显的BMP2和VEGF165 mRNA及其蛋白表达。结论转染BMP2和VEGF165双基因的小鼠骨,髓基质干细胞能同时表达以上两种基因。

Abstract:Objective To evaluate the simultaneity expression efficacy of mouse bone marrow stromal cells (mMSCs) transfected with bone morphogenetic protein 2 (BMP2) and vascular endothelial growth factor 165 (VEGF165) genes. Methods The mMSCs were transfected with pIRES-BMP2- VEGF165 by liposome-mediated method. The co-expression of BMP2 and VEGF165 genes were determined by RT-PCR and immunohistochemical analysis. ResultsRT-PCR and immunohistochemical analysis showed that these transfected cells indeed could produce the mRNA and proteins of the BMP2 and VEGF165 ex vivo and in vivo. Conclusion The mMSCs transfected with BMP2 and VEGF165 :genes will be expression simultaneously of mouse marrow stromal cells.
