天花粉蛋白对荷人肝癌裸小鼠的抑癌作用

摘    要:目的 探讨天花粉蛋白(trichosanthin,TCS)对荷人肝癌裸小鼠的抑癌作用。方法 采用人肝癌原位移植模型。将24只裸小鼠分为TCSⅠ组、TCSⅡ组、TCSⅢ组和对照组,TCS剂量分别为0.2mg/kg、0.4mg/kg、0.6mg/kg,对照组用生理盐水0.2ml/只腹腔注射,共10次。第21天称瘤重和裸小鼠体重,计算抑癌率。应用免疫组织化学技术,检测瘤组织中Ki67的表达。结果 对照组、TCSⅠ组、TCSⅡ组和TCSⅢ组平均瘤重分别为0.99g、0.81g、0.22g和0.20g。用药组瘤重与对照组相比有显著性差异(P<0.05)。TCSⅠ组、TCSⅡ组和TCSⅢ组抑癌率分别为18.18%、77.78%、79.80%。对照组、TCSⅠ组、TCSⅡ组和TCSⅢ组平均鼠重增加为2.22g、1.22g、0.28g、一2.37g。用药组与对照组鼠重变化有显著性差异(P<0.05)。对照组、TCSⅠ组、TCSⅡ组和TCSⅢ组Ki67蛋白阳性表达百分率分别为31.33%、29.92%、10.25%和9.67%。TCSⅠ组Ki67百分率与对照组相比无显著性差异(P>0.05)。TCSⅡ组和TCSⅢ组Ki67百分率与对照组相比有显著性差异(P<0.05)。结论 TCS对荷人肝癌裸小鼠的移植瘤有明显的抑癌作用。TCS抑制肿瘤细胞增殖可能是其抑癌的机制之一。

Abstract:ObjectiveTo investigate the tumor-inhibitory effects of trichosanthin (TCS) on human hepatocellular carcinoma (HCC) in nude mice. MethodsAfter the nude mice bearing LCI-D20 human HCC were treated with TCS at 0.2 mg/kg, 0.4 mg/kg and 0.6 mg/kg and normal saline, the tumor weight, tumor-inhibition rate and body weight of mice were observed. Immunohistochemistry was employed to detect Ki67 antigen expression. ResultsThe tumor weight of xenografts was 0.99 g, 0.81 g and 0.20 g in the control group, TCS I group, TCS II group and TCS III group, respectively. There was significant difference between the control group and 3 TCS groups (P<0.05). The increase in body weight of mice was markedly different between the control group and 3 TCS groups (P<0.05). The percentage of positive expression in TCS I group was not remarkably different from that in the control (P>0.05). However, there was marked difference in the percentage between the control group and other 2 TCS groups (P<0.05). ConclusionsAntihepatocellular carcinoma effects of TCS has been confirmed. Inhibition of proliferation of tumor cells by TCS might be related to its effects in inhibiting human hepatocellular carcinoma.
