摘    要:目的研究丹参素对过氧化氢(H2O2)所致人脐静脉内皮细胞(ECV-304)损伤的保护作用。方法体外培养ECV-304,用H2O2孵育细胞,建立氧化诱导内皮细胞损伤和凋亡模型。用此模型筛选出浓度适合的丹参素。应用噻唑蓝(MTT)法检测内皮细胞生长和增殖活性;比色法测定细胞培养液中超氧化物歧化酶(SOD)活性及细胞内丙二醛(MDA)含量。结果丹参素可明显减轻H2O2所致的ECV-304细胞损伤,中、高浓度的丹参素均可使MDA含量明显下降,SOD活性明显升高,与模型组比较均有显著性差异(P均<0.01)。结论丹参素对H2O2所致ECV-304损伤具有明显的保护作用,其机制与减少MDA生成,提高SOD活性有关。

Abstract:Objective It is to study the protective effects of tanshinol on human umbilical vein endothelial cells(ECV-304) injured by hydrogen peroxide.Methods The ECV-304 was cultivated in vitro.The cells were incubated with H2O2.The oxidative injured endothelial cell and apoptosis models were established.Suitable concentration of tanshinol was sieved from above models.The growth and proliferous activity of endothelial cell were detected with MTT method.The activity of superoxide dismutase(SOD) in cell culture soluti...
