豚鼠空肠吻合术后ICCs的再生及细胞网络再形成研究

摘    要:目的探讨豚鼠空肠壁内ICCs再生及细胞网络重新形成过程.方法利用手术切断的方法,制造ICCs损伤丢失、网络断裂动物模型,冰冻切片和全层铺片免疫细胞化学染色观察.结果术后30~50 d,断端KIT阳性细胞开始增多;术后70 d左右,KIT阳性细胞在断端形成以正常肠壁为底面,顶端向吻合部位的三角形增生区域,在该区域中部KIT阳性细胞的数量较多,且排列不规则,呈团状,突起较短,胞浆丰富,细胞核较大;而靠近三角形底部的KIT阳性细胞逐渐与肌间神经丛周围和深肌丛附近的ICCs相延续.在术后6个月,近端和远端的肠壁已逐渐恢复延续性,肠壁各层结构基本正常,此时KIT阳性细胞亦趋于恢复正常细胞网络分布模式.结论当肠壁内ICCs及其网络损伤后,ICCs可通过再生恢复正常的细胞网络和分布模式.

Abstract:Objective To investigate the regeneration of interstitial cells of Cajal (ICCs) and the restoration of the cellular network after reanastomosis of the small intestine in guinea-pigs. Methods Animal model was established by semi-transection and reanastomosis. ICCs were observed by immunocytochemical staining. Results During the operative period from 30 to 50 d, KIT-positive cells began to increase in the reanastomosed ends. At 70 d after operation, the number of KIT-positive cells adjacent to the reanastomosed ends increased significantly. These ICCs in the regenerated region showed a triangle-like structure in the reanastomosed ends. In the middle of the triangle-like region, many KIT-positive cells characterized by two to three short processes and enriched cytoplasm irregularly organized into groups whereas in the vicinity of the base, a large number of KIT-positive cells tended to connect with IC-MY and IC-DMP. At 6 months after operation, the microstructure of the intestinal wall and the distribution of KIT-positive cells in the region of the reanastomosed ends appeared to be similar to those of the control. Conclusion ICCs in adult guinea-pigs could regenerate and restore their normal distribution and cellular network in the gut after ICCs are damaged.
