日本薯蓣茎叶的显微鉴别研究

摘    要:目的:通过观察日本薯蓣茎叶的显微构造及显微化学反应,明确其鉴别特征,为制定日本薯蓣的质量标准及开发利用提供实验依据。方法取植株的茎叶进行徒手切片,采用显微成像技术进行粉末鉴定;将切片用三氯化锑的浓盐酸溶液或三氯化锑的高氯酸溶液显色,进行显微化学定位研究。结果明确了日本薯蓣茎和叶的显微特征;通过其显微化学反应可知,茎叶部分薯蓣皂苷含量甚微或没有。结论显微特征可作为日本薯蓣的鉴别手段;显微化学方法及其反应现象作为日本薯蓣资源评价以及筛选育种过程中一个快速选择的检测指标,是切实可行的。

Abstract:Objective By observing the microscopic structure and microchemical reaction of stems and leaves of Dioscorea japonica, a clear identification was obtained, providing a scientific ba-sis for the further developm ent of Dioscorea japonica quality standards and exploitation. Methods Freehanding the plant stems and leaves with powder identification by microscopic im aging techniques, then coloring the sections with concentrated hydrochloric acid solution of antim onous chloride or per-chloric acid solution of antim onous chloride, we conducted a histochemical localization study. Results Clear the microscopic characteristics of D. japonica stems and leaves and by their microchemical study, little even no dioscin exists. Conclusion These microscopic characteristics can be as a means of identification of D. japonica; Microchemical method and reaction phenomena as a quick select in-dexes of D. japonica resource evaluation and selection breeding process, is easible.
