黄精煎液对衰老小鼠组织端粒酶活性的影响

摘    要:目的 探讨黄精煎液对衰老小鼠组织端粒酶活性的影响。方法 采用端粒重复扩增-微孔板杂交法检测衰老动物组织黄精治疗前后的端粒酶活性;采用改良TBA荧光法测定组织丙二醛(MDA)的含量。结果 黄精治疗后实验动物脑、性腺组织端粒酶活性明显升高,而其MDA含量无显著变化,MDA含量与端粒酶活性无相关性。结论 黄精可显著升高衰老动物脑和性腺组织的端粒酶活性。

Abstract:Objective To investigate the effect of ploygonatum sibiricum (P.sibiricum) on telomerase activity in animal tissue and the mechanism.Methods The telomerase activity in aging mice tissue before and after treatment with P. sibiricum were measured by TRAT microtiter plate hybridization method and tissue MDA was measured by modified TBA fluorescence method.Results The telomerase activity in gonad and brain in the experimental animals was significantly increased after treatment with P.sibiricum, but the MDA levels had no significant change. There was no relationship between tissue MDA levels and telomerase activity.Conclusion P.sibiricum can significantly activate the telomerase activity in brain and gonad tissue in the aging animals.
