线粒体损伤在散发性Rett综合征发病中的作用

摘    要:目的探讨线粒体损伤在Rett综合征(RTT)发病中的作用.方法采用细胞融合技术将6例Rett综合征患儿及9例正常对照血小板与无线粒体DNA的ρ°细胞融合,用极谱法评价融合细胞氧自由基水平,用流式细胞计测定细胞凋亡率,应用电镜和TUNEL观察细胞凋亡.组间比较采用t检验.结果RTT患儿组抗氰呼吸较正常对照组增高23%(t=4.76,P<0.01),应用100μmol/L的H2O2刺激融合细胞6h后,细胞凋亡率正常组为(3.6±1.1)%,RTT患儿组为(9.9±2.7)%,显著高于正常对照组(t=6.30,P<0.01),电镜和TUNEL均证实凋亡.结论线粒体DNA转移后RTT患者融合细胞线粒体呼吸链氧自由基水平增多,RTT融合细胞对过多氧自由基的凋亡易感性增高,线粒体损伤在RTT发病中可能起着一定作用.

Abstract:OBJECTIVE: To investigate the role of mitochondrial lesion in children with Rett syndrome (RTT). METHODS: The platelets from 6 cases with Rett syndrome and 9 normal controls were fused with mitochondrial DNA-lacking rho degrees cell by polyethylene glycol-mediated fusion technique. The oxygen free radical was evaluated by the polarography with substrates of vitamine C and TMPD (N, N, N', N'-tetramethyl-p-phenlene diamine). The rate of apoptosis was determinated by flow cytometry. The apoptosis was observed by electronic microscope and TUNEL method. The t test between groups was adopted in study. RESULTS: In RTT patients, the anticyano-respiration significantly increased by 23% (t = 4.76, P < 0.01). After 6-hour coincubation with 100 micromol/L H(2)O(2), the rate of apoptosis is (3.6 +/- 1.1) % in normal control, and (9.9 +/- 2.7) % in RTT group with significant difference (t = 6.30, P < 0.01), the existence of apoptosis was confirmed by electronic microscope and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick-end-labeling (TUNEL). CONCLUSION: After mitochondrial DNA transfer, the oxygen free radical increased in mitochondrial respiratory chain in cybrids of RTT children. RTT cybrid cell lines demonstrated an increased sensitivity to H(2)O(2)-induced apoptotic cell death as compared to control cybrids. The mitochondrial lesion might play a role in the pathogenesis of Rett syndrome.
