Expression of Ku80 gene in human lung adenocarcinoma A549 cells before and after exposure to irradiation

Abstract:Objective To investigate the changes of Ku80 gene expression in human lung adenocarcinoma cell line A549 before and after radiotherapy. Methods Human lung adenocarcinoma A549 cells were inoculated to nude mice and exposed to different doses of irradiation. The expressions of Ku80 protein and mRNA in A549 cells were detected by immunohistochemistry and real-time Q-PCR, respectively. Results The expressions of Ku80 protein and Ku80 mRNA in A549 cells were significantly increased 24 and 48h after exposure to 1Gy, 4Gy and 8Gy irradiation (P〈0.05). After cells were exposed to 4Gy and 8Gy irradiation for 48h, the Ku80 protein expression was higher than that exposed with the same dose for 24h (P〈0.05). Conclusion The expressions of Ku80 protein and Ku80 mRNA in human lung adenocarcinoma A549 cells are increased after exposure to irradiation in a dose-and time-dependent manner, which suggests that Ku80 gene expression may be involved in the repairing of radiation damages.
